i am applying compression into the transwell insert and this compression cup occupies all the surface area of insert thus i can only add minimal volume into the transwell insert, most media is ejected out after applying compression.
my question is, if enough media is placed at bottom of 6 well plate, can media liquid seep upwards and into transwell insert, preventing drying of cells?

your question needs more clarification, that mean how much media you add to the top of the insert and how much you add in the well in 6well plat. if I understand from your question the insert with 1.5-2ml and the well with the same amount the cells will be happy and they will be not dry. you need to add the media to the insert first and then add the media to the well.

if no media is placed into transwell, only present in bottom well, media cannot enter the transwell? my compression device that puts in a compression cup into the transwell insert ejects most media. i was hoping if there is media at bottom plate, it can precent drying of cell
