At our lab we are currently struggling with bad X. laevis oocyte viability, fragile membranes, soft and deformed cells. In TEV recordings, the membrane potential of many cells is often less than -10mV. We see this especially in oocytes from the latest batch of frogs, which are supplied by nasco. Other oocytes that we sometimes borrow from another lab seem to be much better, therefore we think that our protocol, for defolliculation, maintenance, RNA injection etc., is fine. We initially suspected an infectious cause, but a vet examination and bacteriology didn't confirm this.

Does anybody have experience if maybe there might be oocyte quality differences between different X. laevis frog suppliers? Do you have any suggestions for good european suppliers? Is anybody maybe even breeding in their own facility, and would be able to share some resources for doing this?

Many thanks!
