Currently working with THLE2 cells but don't have any experience with these cells. Do they only grow in BEBM media or they would grow well in DMEM as well?
I've not worked with these cells before but there are basics to cell culture that can be applied to your question...
Normally the culture collection that you obtained the cells from will indicate what growth conditions are. ATCC states BEGM.
Although most cells could grow in DMEM, DMEM comes in different makeups - high/low/no glucose, high/low/no glutamine, with/without sodium pyruvate, with/without sodium bicarb. You could try matching the ingredients that make up BEGM but then why not just buy BEGM in the first place?
Another consideration is whether the cells are differentiated and if BEGM helps or maintains cellular differentiation. Most likely if you grow the cells in standard DMEM you may lose this differentiation. You also need to consider whether using DMEM to BEGM will cause expression of different genes or change the morphology/function of the cell line.
If you still want to press ahead, just split a spare flask and add in DMEM to see what happens!
I am planning to work on THLE-2 cells and I have the necesasary media for it. My question is: Did you pre-coat your flasks like ATCC recommended or not ?
