Basically, I am looking at design where I have exogenous Sequence A- Intein N and endogenous Sequence B - Intein C - Endogenous sequence C.
Would this strategy work, if I had to obtain the sequence Endogenous sequence A - Endogenous sequence C ligation ?
In principle that should work. If all other criteria are fulfilled, the additional Sequence B should not interfere with the splicing reaction. Cheers Tobias
I agree with Tobias - this should work. You should take some care with the design of the link between sequence B and the C-intein so that sequence B will not interfere with the intein folding.
Indeed, a good number of natural inteins have an additional domain inserted into the intein sequence, which is also excised during intein removal.
