Can anyone please give a working protocol for Taxol washout experiment, I am trying to study the impact of MAPs on Microtubule stability. I am using flow-chamber, attaching Taxol (1uM) stabilized microtubules to the coated antibodies in there, washing with 10 volume of BRB80 without Taxol. I do not see much depolymerization in the Microtubules, most of them remain intact, hardly 1% undergoes catastrophe.
