Hello researchers,
I am trying to do an imaging study with PI for the live/dead cell assay in HeLa cells infected with virus and incubated with nanoparticles as well. Can anyone please share the working protocol for the PI staining, please?
Also, I am thinking to stain the cells with Hoechst as a control. Please share your thoughts.

I think the standard procedure should have worked. We generally follow these steps and used to get fluorescence with breast cancer cells.
-> Fix the cells with 90% ethanol in ice for 15min
-> Wash with PBS
-> Add the staining solution and incubate it on ice for 1hr in dark
Staining solution: Propidium Iodide (1mg/ml) and RNAse (1mg/ml) & Triton -X (0.1%) in its PBS
PI and RNAse should be freshly prepared.
