I am isolates Photobionts from lichens and cultured them. I am getting sufficient biomass on agar plate containing 3N BBM media and broth as well. How can i preserve this biomass for future studies?
Two ideas come quickly to mind - does anyone at Central University of Punjab have cryopreservation equipment, as this is an excellent way in which to preserve . (You will find methodologies in the literature) Or, if you have access to controlled temperature incubators with light sources, you can reduce the temperature at which both your broth cultures and plates are kept, slowing growth and reducing the need to subculture. You can probably use Lugol's solution - iodine in potassium iodine. Algal samples for morphological studies are preserved very well for long term storage using this.
You can use DMSO 10% final concentration or even 15% final concentration for larger cells. (15-20 minutes).
This fixation doesn't change the optical properties of the cells, opposite to the paraformaldehyde or glutaraldehyde when visualize them in the microscope.
