I'm working with a DNA purification kit in order to purify and collect plasmid DNA from E. coli bacteria. The manufacturer's instructions say to resuspend a bacterial pellet formed after centrifugation in their special resuspension buffer. Do bacteria truly need to be resuspended in a special buffer in order to prevent osmotic shock, or can they just be resuspended in distilled water?
