I'm working with a DNA purification kit in order to purify and collect plasmid DNA from E. coli bacteria. The manufacturer's instructions say to resuspend a bacterial pellet formed after centrifugation in their special resuspension buffer. Do bacteria truly need to be resuspended in a special buffer in order to prevent osmotic shock, or can they just be resuspended in distilled water?
They are likely to undergo some level of osmotic shock and quite possibly a significant amount of lysis. I would suggest using the appropriate resuspension buffer.

Yes they would undergo osmotic shock. It is always advised that we use special buffer for vitalization. The reason is that the distilled water would definitely be a region of lower concentration and as much the organism would lose everything via osmosis.
