I am extracting DNA from blood, and due to waiting for buffer solutions to arrive, the blood samples appear to have dried in their plates. Some were clotted, so have had EDTA added to them also. There is 20ul in each sample. They have been in a 4 degree fridge for the last week, and the necessary buffers should arrive any day. I would just like to know if anyone has had something similar, or if there's anything I need to do next? I will be using the DNEasy 96 QIAGEN kit - the blood samples are currently in the well-plates.

Yes, It is possible to extract DNA from a dried up sample. Traditionally the DNA was extracted from dried up blood sample on a filter paper. If you had kept the plate at 4 degree C, it should be fine. In fact, the dried blood samples do not have the same time and temperature constraints as wet. No refrigeration is needed. However, you may have to follow some laborious protocols and may be multiple extraction steps. The longevity of the dried up sample could be up to several months. People who extracted the DNA found that there was no degradation of DNA and increased temperature or humidity did not have any adverse effects. Although, these experiences are not mine but I am writing to you about other investigators who had to face this problem. Please use your own discretion.

Thank you so much for your response. That's good to know. I will have to remain with a certain protocol, in which PBS and other buffer solutions are added to the samples (they should be 20 microlitres each of liquid blood that lyses with the buffers), so I just hope there is a way in which the blood re-susoends when in the centrifuge, but this is great to know that my samples won't have degraded.
