I have some hybridomas that were generated for us commercially and I do not have too many details on what mice or myeloma was used to generate them. We would like to adapt them to CD Hybridoma medium and to be safe I could add the 250X cholesterol supplement from Thermo. What I need to know is if this will harm hybridomas that do not require cholesterol for growth. Thanks for any feedback.
Followup 4/20/17: I did manage to contact the company that made the hybridomas and found out that they are NS2 derived. Their standard protocol uses IMDM with 20% FBS and HT supplementation or HAT selection for propagation. I plan on trying to grow a fairly large amount of cells and then transferring them into CD Hybridoma medium for antibody production in one shot. Since we're not trying to make more than a couple of mg of MAb at a time I hope that this will work.
