I was trying to adapt BHK-21(C-13) cells to serum free conditions. Cells were initially cultured with 10% FBS and I was trying to intervene the serum. For this I used Excel CD 302 medium, VP-SFM medium using the sequential adaptation method. Upto1% FBS cells were growing fine different media and cells were not growing in the media with less than 1% FBS. Can any one suggest me the why these cells were not growing in serum free medium? Any supplements required for BHK 21 cells to grow under serum free conditions?
It seems very interesting observation for me. I recently have found that adult serum contains titin/connectin.
These another authors indicated the differentiation in BHK cells.

I do not understand why fetal calf serum (FCS) is used generally at this time.
Therefore, I consider that some amount of connenctin/titin, an adult marker potein, might be contaminated in your purchased FCS.

As mentioned in serum titin/conenctin will be present in trace amounts. Is these proteins are required for the growth of BHK-21 cells. I would like to adapt these cells to grow under serum free conditions. Is there any method or media to grow these cells under serum free conditions?
serum-free media is used to culture continuous cell lines which are already adapted to serum-free culture. These include the degree of chemical definition, e.g. serum-free (SF), animal-derived component free (ADCF) or chemically defined (CD), and the type of medium, e.g. basal media, medium supplements, or full replacement media. It is very hard to adapt BHK-21(C-13) cells to serum free conditions because dividing cells require metabolites such as carbohydrates, lipid, and proteins for formation of membrane structures, chromosomes, cyto-skelatal elements. Despite all efforts only supplementation of sterols can provide them some strength. alternatively amino acids an glucose are give as supplements in serum-free media to adapt cell lines.

We tried various commercially available serum free media like EX-CELL 302 serum-free media, CHO-CD Chemically Defined Medium, VP-SFM these media were prepared as per the instructions given by the manufacturer. Other than that we didn't add any supplements. Can you recommend any specific supplements? These media were used to culture CHO cells under serum free conditions and were growing fine. For BHK are there any specific supplements required?
You can try Merck Miilipore Cellvento SAF (BHK) 200 and Gıbco CD BHK 21 Production serum free medium. These serum free media are produced especially for BHK 21 cells.
