There is less chance of changing if you re calculate the fluent file as i know. Are you sure that you didn't change anything before initialize?

I have a doubt regarding your grid independence.

Also this may happen if your initialization and boundary condition if you are giving for the same point are mismatching.

Change in value is too much so this is not from simple calculation error propagating.
