Hi, we have Shimadzu UV 2600i UV-Vis-NIR spectrophotometer in our lab. But when we are trying to get the absorbance of Glass/Mo sample it is showing erronious results though for the baseline, we took glass samples and during measurement glass was the refernce and glass/Mo was the sample to be measured. I have attached the result here. Kinldy guide us for the same.
The error is primarily caused by extremely high absorbance values (up to 11 AU) and improper handling of solid samples. To resolve this:
- Use reflectance mode for solids or reduce sample thickness.
- Ensure measurements stay within the instrument’s photometric and wavelength limits.
- Validate baseline correction and cuvette materials.
I agree with the previous answer concerning a too thick (dark) sample. Besides reducing the sample thickness, check for a good flatness and smoothing of the glass surfaces. An integrating sphere can help very much in case of not completely regular glass surface and parallelism between the two surfaces crossed by light beam. Greetings
I agree with the above two answers. The absorbance peaks are getting saturated maybe due to the thickness of the sample or its not fully transparent which can create error in the measurement. You can use ATR mode for thicker samples.
