During GC/MS analysis of organic tracers in air,we have to derivatize the polar groups of organic compounds.For this,i have chosen the BSFTA+TMCS reagent.I wanna know that now,should i need a solvent pyridine for this or i can do it without pyridine.what is the role of pyridine?
There are various approaches to extract (13)C-labeled sugars (glucose, fructose, maltose, sucrose, myo-inositol as well as glucose from starch) from plant tissues and to analyze their isotopomer distribution by gas chromatography-mass spectrometry (GC-MS). Sugars are derivatized with N,O-bis(trimethylsilyl) trifluoroacetamide (BSTFA) into their Si(CH3)3 derivatives. Electronic and chemical ionizations are used to obtain suitable fragments for metabolic flux analysis. Unique fragments are identified by computer simulation and experimental verification with labeled standards. Linear equations for separating information from glucosyl and fructosyl moieties of sucrose are used.. Finally, mass distributions are corrected for natural isotope abundance using a home-written program.

Since you already have the catalyser, I would say you don't need pyridine but check the application note for further advice on your specific compounds
Yup, both as an aprotic solvent media and catalyst, pyridine helps, and possibly for further solubility of the derivatized compounds ! best wishes.
Pyridine is basic and reacts with the acid produced by the silylation reaction. These helps to drive the reaction. By using a sufficient excess of reagent ther reaction will go fairly rapidly without the pyridine solvent. If you just do not want to use pyridine, THF and acetronitrile have been used as silylation solvents.

In many cases, the need for a solvent is eliminated with silylating reagents. The completion of the derivatization process in silylation is usually observed when a sample readily dissolves in the reagent.
