I am currently measuring glucose concentration (OD value) in MSC (KURABO cell) culture media every day. During this time, I used fresh storage cell culture medium as a standard. I can understand why cell culture glucose concentration decreased during the time. But I cannot understand why the OD value of storage medium (Standard) also decrease. Please help me to clarify this problem.....my storage temperature is 4C, I am using our own cell culture medium.

There are basically two options:
1) the glucose concentration in your 'fresh' medium really decreases over time (maybe due to bacterial contamination),
or 2) the glucose concentration in your 'fresh' medium is stable but your assay procedure is not working properly (giving you falsely low signal).
I don't know what assay you are using. I suggest to use several concentrations of a commercial glucose standard as your reference. Always store this standard in the freezer to avoid degradation of glucose. I am using enzymatic glucose assays with detection of NADPH for years. It's VERY reproducible.
