Most works use the TNBS assay to quantify free amines on the chitosan when making micelles, to quantify the degree of substitution by the hydrophobic moiety (or any further modifications).

I am running into trouble with chitosan oligosaccharide lactate (https://www.sigmaaldrich.com/GB/en/product/aldrich/523682) and quantifying the free amines with ninhydrin. Is this because of the lactic acid occupying the amines? Is there another reason?
