I have used 10% FBS and this image i took after 3 days of passage. and I don't see any cells floating from where i think it might not be contamination but I don't understand what it is. If this is cells are getting lysed, what can be the stress factors.
Yes cells do not look quite healthy. In 3 days they should have formed small colonies at least. The granules in the background indicate dead cell debris. Cells could be under some stress for example how long did you trypsinize the cells & the strength of the trypsin solution used? Also HepG2 cells grow better in DMEM with low glucose (1g/L)
Hi. At what density did you split your cells? After 3 days they should be a lot closer together.
These cells are not dividing and are actively dying. I would discard these cells and start a new batch. Use DMEM with 10% FBS and 1% Penicillin/streptomycin.
