First of all, I appologize my poor English skills.
I am work at biopharm.
We test our NK cell's cytoxicity capacity using IFN-gamma test.
But for the IFN-gamma assay, pre-treatment PMA/IONMYCIN above 4hr incubate.
That is so long time for our job.
Those it can availble shorten incubate time?
We just wait 4 hour before our final product.

It could be because the effector function of NK cell makes use of integrated signaling from an array of activating receptors on NK cells.
The triggering of cytokines and chemokines secretion requires a synergistic combination of several receptors. This then leads to convergence of signals to a central signaling molecule in order to reach a threshold level which is required for activation of NK cells.
So, a minimum of 4 hours of incubation with the stimulant could be necessary to test NK cells cytotoxic capacity.

Just a thought: you can have batches of cells with different hour duration 4, 3.5, 3, 2 and 1 hours and check the cytotoxity capacity. it will give a clear picture.

I will try 2,3,3.5,4 hour treament test.
I forgot before this job, I was researcher.
Actually, I working in QC team, because I have never thought about preliminary test.
