I’m running 0.8% agarose gel and I had two incidence that the diffusion of the samples and ladder showed a line coming down from the well what could be the reason ?
I run the gel at 75V for 45 minutes and I left it more that an hour to solidify
1/ Make sure that your agarose is well-dissolved during the boiling process, without any flakes remaining.
2/ Try lower voltage, e. g. 50 V.
