Hi all

I am doing ELISPOT for the isolated spleen and BM cells of mice to evaluate IgG and IgA Antibody secreting cells against pneumococcus. I immunized the mice earlier. and now at the day of sacrificing after isolation of cells we coculture them in the elispot plates with the same antigen and after 5 hours 37 we add secondary ab and then incubate in 4 overnight and development.

Which we have spots in the control group.

Any suggestion to get ride of this spots will be appreciated.
