Over time the pyruvate in solution will self react and dimerize forming parapyruvate and can also further polymerize. Not sure of the cytotoxicity of that or the color.

I may suggest to keep your solution under Nitrogen or Argon in a tight bottle to prevent the possibilties of this colour changes, which may be due to some sort of polymeriztion.
