When performing Karyotying using Peripheral blood sample, we add 250 microliters (collected blood into heparin tube) in to 5 ml PB-MAX culture media and incubate it for 72 hrs in CO2 incubator. But recently in few cultures, we observed dark brownish red colour while others are pinkish red as usual. And those dark red colour samples do not give good results. (either don't have chromosomal spreads and cells or some times very few spreads are there)

[All those samples are from infants. But not all the infants' cultures give dark red colour.]

What could be the reason?
