Hello.

I am currently running haemolysis assays to measure inhibitory effect of some complement regulatory proteins. The assay involves complement activation by either the classical pathway (CP) or alternative pathway (AP) by incubating sheep/rabbit erythrocytes with normal human serum (NHS).

I want to know as to why sheep erythrocytes are usually used for CP activated (by incubating the sheep erythrocytes with ambocepor which is a rabbit anti-sheep antibody, addition of NHS) haemolysis assays while rabbit erythrocytes are preferred in AP activated (addition of EGTA to inhibit classical pathway activation, addition of NHS) haemolysis assays.

Can sheep erythrocytes be used for the AP activated haemolysis assay aswell? adding EGTA aswell to inhibit CP activation?
