I came across klenow fragment when I was reading about HiTS-FLIP method. I know klenow fragment doesn't posses 5'->3" exonuclease activity but what advantage does it provide for ?
unlike the thermostable polymerases Klenow is very high fidelity at low temperature (37c). It may be that in order to use the technique on existing equipment it is more convenient to use low temperatures. Also possible is that it is used to study dna/protein binding at biologically normal temperatures while many other polymerases will dissociate the bound fraction at their optimum extension temperatures.
