Why do we set up primary and secondary culture of pichia pastoris to make electro competent cells?

I am unable to get the point or principle behind secondary culture from primary culture of Pichia pastoris KM71H ?

Hi there,

What you actually need to prepare competent yeast cells is an actively growing culture at OD between 0.8 and 1. It could be primary culture if not overgrown otherwise you need to dilute it and grow it as a secondary culture until the right OD.
