I am trying to ascertain the Fibrinolytic activity of 3 of my proteins by zymography. One of the proteins is a low Molecular weight protein while the other two are about 25-30kDa in size. However, digestion was observed at the top, just under the stacking gel in case of each. Also, a continuous white line of digestion was observed along the dye front. I used a 15% native gel. I am finding it difficult to interpret the zymogram, any help would be appreciated.
Hi Chitra Nair could you post a picture of your zymogram?
High salt (and/or organic solvents) can retard protein mobility in SDS PAGE. Another issue could be the gel itself.
The protein band was extremely faint to photograph, it is the one in well.no. 2 from the left, sandwiched between the two white lines.
