The paraffin slides I got have a lot of bubbles (as in attached photos – w4, w5, w6). The samples were fixed with 10% neutral buffered formalin for 1 day, 1 week and 2 weeks. Then were decalcificated with 10%EDTA at 37°C for 10-20 hours.

I tried to cut paraffin slides with both 4um and 10um thickness. Some of them were oven baking for 4hours or some were baking for 22 hours.

I also attached a photo of RmW1-8 I got last year. This sample were decalcificated first then processed the fixation. According to literatures, we need to do fixation first before decalcification. That's why i changed the protocol. However, using both protocols, the bone slides have many bubbles.

Just wondering whether you have any suggestions and thoughts on it.
