Look lime a bacterial contamination to me but the particles are bit bigger.

Do you see any turbidity in the medium? and what us the color of the medium?

Are you using antibiotics in the culture?

Yes Sir I am using Pen-Strep with Amphotericin B.And theres no turbidity in the medium,nor any colour(pH) change

Are these from the same microscope? The microscopes in my institute in the Main Research Lab and in the Extension Research Lab are different brands and only one has the Phase Contrast setting, so my cells (the same T25 flask) looks different from one microscope to another.

I would suggest you asking the other lab about the components in the growth medium that they used for PANC-1 cells. Do they use 5% FBS, or 10% FBS, or 20% FBS? How they passage their cells - do they neutralize by centrifugation before plating? And their rules and regulation of cell culture - what are the Do's and the Don't's that they apply for culturing PANC-1 cells.

Some indications of bacterial contamination are notable where the space between cells appears uniformly granular and have a shimmer. Some contaminants are clearly visible by eye, changing the color , turbidity of media and when cells are not attached to the plate but are floating in the media.

Most cells with ageing lose intracellular material. It is notable with RCBCs.

The scientists using the toxic iron containing ~50nm nanoparticles conjugated with an antibody, say CD34, such as Miltenyi particles-CD34mAb to isolate cells- there is evidence from transmission electron micros copy of cells after 24,48,72 hours that the Milteny get internalised and remain trapped- they are apparently not biodegradable that this multi billion company promoting the expensive products appears to claim- have provided no evidence published - or sent to the FDA for approval that they procured and have not modified since the publication of the evidence in the video clip- see link on YouTube below. The video clip also provides evidence form confocal laser microscopy surface scannig that the Miltenyi particles get internalised and fused with vesicles.

You can try to take a small aliquot of the medium from your culture and "seed" in medium with 15%-20% FBS. If there is some kind of contamination, you should see outgrowth of this particles.

I would like to suggest using "inactivated" serum (FBS) and then give it a try.
