I generated S. aureus mutants by transduction and then complemented the mutants with the wild type alleles using a shuttle recombinant vector. the problem is when I done broth micro-dilution assays I got MIC values higher than the the wild type. Ideally I should expect the same or/ lower value than the wild type. Any idea.
