I have been extracting RNA from oat spikelets using the Sigma Spectrum Plant Total RNA Kit. I am getting super nice yields, and my A260/280 ratios are between 2.1-2.2. However, my A260/230 ratios are a bit higher than expected - around 2.4. A good ratio is around 2-2.2, and a ratio lower than that would indicate carry-over of e.g. guanidine or phenols. But what does a high ratio indicate, and could it be problematic when using the samples for RNA-Seq? Samples look fine when run on a gel.
