So I've got a text file. In it a have a lot of nodal values S11, S22,S33.... . I am not sure how to get a single value (for each S11, S22, S33.... )of normal and shear stresses to be able to get the global stiffness matrix of the specimen?

Stresses are calculated at the integration points. Use element stresses and you will get the for each integration point the stresses you need. The stresses at the nodes are extrapolated to the nodes and / or interpolated between elements.

No, I mean from the output data from abaqus (Stiffness values are in one column in txt or csv file) making from it a normal 6x6 stiffness matrix for specific material? Most of the people as far as I have seen use matlab, the question is how?
