I did a western blot on fresh heart tissue lysate from adult wister rat. Lysates generated form the ventricles gave a triplet band corresponding to the three phosphorylation states of the protein (P0, P1, P2), but lysates generated from the atria and septum gave a doublet (P0 and P1) or single band. However, I know that Cx43 undergoes a lot of post translational modifications, but a search of the literature for the reason for different number of bands from different tissues of the same animal was not successful. Please can anybody explain why?
