I am going to study the effect of a hypoglycemic drug on endothelial progenitor cell (EPC) functions. The published work has reported that a 24h 25~33mM glucose treatment is enough for impaired EPC viability, migration and tube formation ability (PMID: 35882957, 28799229). In my study, cells in control group were cultured in EGM-2 culture medium supplemented with 10% FBS (Lonza) and 1% antibiotics. For high glucose group, the cells were treated with 40mM glucose. To my surprise, CCK-8 shows that the high glucose group have higher viability after 24h or 48h treatment. The cells were treated with 40mM glucose for 24h or 72h and resuspended to perform tube formation assay. High glucose group also performed better. The passage of the cells used in tube formation and CCK-8 experiment were 3-7.
I am not sure the suitable culture condition to mimic the high-sugar environment of diabetes for EPCs and wonder if it because high glucose promotes differentiation of the cells. Can you shed some light on this or give some suggestions on experimental conditions optimization? I would appreciate it~
