Hello everyone. We are doing quenching analysis of Fluorescein and DABCYL at low concentrations (1x10^-6 M of fluorescein) in DMSO. Keeping Fluorescein concentration constant, DABCYL concentration is increased and the Fluorescence spectra is recorded. We have noticed that when we added from 0.02 to 0.1 equivalents of DABCYL per equivalents of Fluorescein, the fluorescence increases. Later, when adding over 0.1 equivalents of DABCYL the fluorescence decreases (as expected). We have not observed this behavior at larger concentrations of fluorescein and DABCYL (1x10^-5 M of fluorescein). Do any of you could help me to explain these observations?
