I have been propagating a poplar hybrid (717) in sterile tissue culture with 1/2 MS media and no hormones. My last few batches haven't been rooting well. I have replanted some into 1/2 MS with IBA, but there are still no roots. I welcome any suggestions you might have!

I suggest that you sub-culture the shoots on MS medium containing 0.2 mg/L IAA or 0.1 mg/ NAA.

Some woody plants are very difficult in TC. All colegues from research gate are right. You have to chose a type of auxin at good concentration. Did you putt microshoots on medium with auxin for whole rootings stage? Auxin is neccessary for formation roots primordia and later can inhibit roots elongation. Which medium do you use for multiplication. Sometimes the cytokinin used during regeneration stage inhibit formation of roots primordia. Is your poplar hybrid rich in organic coumponds?

Thank you very much for all of your suggestions. I am now trying multiple combinations of IAA and IBA, as well as darkness regimes. I am still having trouble rooting my poplar hybrid.

However, my last few batches have not had the best rooting percentage (only about 2 in 18 would develop roots). Could it be that the plants are getting old or tired?
Dear Ahmed, I am going to try adding activated charcoal to my media next. However, I am worried about not seeing the root development if it makes the media too dark. Have you run into this issue previously?

Switching from full strength multiplication medium to half MS base rooting medium might change the water potential. Water potential play an important role for mineral absorption. Use full strength MS base medium supplement a very low concentration of IBA and NAA separately and in combination for confirm rooting.

If you explants are not rooting, but have a well-developed shoots mean that shoot-derived auxin does not transported properly. IBA/IAA is not very effective in this case, because you need good auxin transport, not auxin itself.
MS has a very high N contents what is not beneficial for the rooting (as well as other process). And far from optimal ions ratio.
Try to use better medium like Hg or more close to natural.

Thank you very much for all of your suggestions. I have ended up using a mixture of activated charcoal and IBA for 2 weeks. This produced strong roots in my poplar hybrids. I then transferred them to 1/2 strength MS medium. This seems to have worked as all my cuttings have produced roots!
