I am encountering some problems with my HUVEC culture. After the third passage, the cells are dying, and I have no idea what's wrong. I am using F12 medium and typically seed approximately 1 x 10^5 cells. I suspect the issue may be related to the cell concentration.
I would greatly appreciate your insights on this matter.

Hello Julia,
I feel the issue is the growth media. You will have to use DMEM:F12 (DMEM/F-12 is a 1:1 mixture of DMEM and Ham's F-12). More information on the composition of the culture media for HUVECs may be obtained from the link provided below.

I suggest to be carefull with HUVEC.
You can either find medium ready to use with PromoCell or other suppliers.
If you want to create a home made medium, typically you will need to supplement with heparin, growth factors...

in our lab, we grow HUVEC in M199 with 20%FBS + endothelial cell growth supplement +heparin and it works fine
