I would like to extract total protein from Scenedesmus. Some how even from 1 g wet biomass very very low yield of protein (1 ug/ul). IN bacterial we use to get so much of protein from 3 ml culture (50 ug/ul). Could any one please explain why low yield? how to overcome this problem. Or we should start with one liter culture?
basically, the more cells you have the more proteins you get. however, I would use RIPA buffer for lysate cells if you have problems with SDS buffer

Well well, welcome to the wonderful world of protein extraction from microalgae!
Unfortunately, extract proteins from microalgae is not as easy as from E.coli.
You must use some weird protocols including elevated temperatures, NaOH, long time of extraction step (up to 1h30min) to have the chance to get protein extract at mostly 0.05-0.1 ug/uL ! I joined a 1981 paper on that topic.

What make this algae so rigid not to release the proteins? Is it tough cell wall? or lysis buffer composition. As Dr. Massar Alsamraae pointed out?
If these the case, how about the digestibility (whole cell based feeds) ?
