I am trying to streak the K-12 MG1655 strain on LB-Agar plates and there is a lot of heterogeneity in the size of the colonies. When I pick up 1 discrete colony and transform it, even after spreading the colonies show heterogeneity. What could be possibly wrong?
What you are seeing on your plate looks entirely normal. I think the different sizes are primarily explained by colony density differences, a colony in a clear area does not have local competition for nutrients which in a dense area it does. I think you would find more homogeneity (but not total uniformity) if plated at a much lower density.

I found from the literature that they are very fast-growing and reduced incubation time. It resulted in more uniform-sized colonies.
