In my case I use 3D Bioprinting method and I use the same amount: 2500 MB231 and 2500 MCF cells/well, just add the same amount (2500 human fibroblasts) to make nice spheroids. Otherwise, spheroids may be loose or do not form at all, especially in MB231 case. MB231 spheroids always grow bigger (up to 1 mm), while MCF-7 spheroids usually are much more compact and do not grow very big, up to 250-350 um in diameter. I think, people use larger amount of MCF-7 cells just to make similar spheroids to compare the tumor microenvironment, as it depends on oxygen and nutrient gradient. In more hypoxic spheroids it could be very big necrosis in the center.
These two cell lines (MCF-7 and MDA-MB-231) have different biological features and behavior (they represent differemnt types of human breast cancer, express differenmt markers, possess diffrenet growth and invasive characteristics). And we need to optimize the 3D cell culture conditions of each individual cell line .
