I am trying to establish ρ0 cell in MCF-7, PC3 and MKN45 cell lines.

The cells are cultured in DMEM high glucose supplemented with uridine, pyruvate and 50 ng ethidium bromide. After more that 40 days, the PCR results did not show the ρ0 cell establishment.

Is there a protocol to select the mitochondria depleted cells from the another cells in culture?
