It is reported in many studies that micro-RNA's (mi-RNA's) blocks mRNA translation by binding to its 3'UTR, and repressed mRNA is stored and destroy in cytosolic P bodies. So i wish to know which technique will stain accumulation of that repressed mRNA in P-bodies or how we can identify which mRNA is accumulated in P bodies?
You probably can't specifically stain repressed RNAs. You can perform RNA FISH to look at a specific mRNA and co-stain with a p-body marker like GW182 and look for colocalization. To identify specific mRNAs, you should perform iCLIP with a p-body protein that binds RNA.
Dear Mark Livingstone sir, i read in one review article that insitu technique was used to stain repressed mRNA in P bodies, but they wasn't explain specific technique and not replying as well, and GW182 found to degrade all repressed mRNA by deadenylation so how i will identify that they are destroying specific mRNA?
