I have performed the disc diffusion assay against the plant material extracted in four solvents (Methanol,Acetone, chloroform, pet ether) against six bacterial strains and i used four different concentrations i.e 100mg/ml, 50, 25, 12.5mg/ml. now i want to ask how can i apply 3 way anova or t-test or someother test as my data is somewhat of this type...
Hello Lalita, I have seen your data and I think this is factorial experimental design where the factors (1) Plant extract (at 4 levels corresponding to solvent employed), (2) Extract concentrations (at 5 levels including the positive control) and (3) Test isolates (at 5 levels corresponding to the number of isolates employed for the test). You would have to establish main effects and the interactive effects which will be produced as an output of your analysis. You could use SPSS or Minitab or Design Expert as your software. If you like you could also do a univariate analysis which suits your data well because it analysis data with only one dependent variable but with any number of independent variables/predictors. The only dependent variable i see in your data is zone diameter. I have neither seen nor adopted a three-way ANOVA before so i cannot help you there. Whatever you do, try to keep things simple to facilitate reproducibility of your research. Good luck.
