I have prepared carp gill lamella using H&E and observed the number of vacuolated cell were increased following intoxication. I decided to identify whether that's really mucous cell or not.
I think AB-PAS is a good. Please let me know if you have any experience for staining of gill lamella to observe mucous cells.
I have used H & E but also tried Pollack'sTrichrome stains and even Alcian Yellow. There is some research out there that suggests you can determine the type of mucin using stains - a fish researcher by the name of Pickering has done a lot of work on this - try this publication: http://link.springer.com/article/10.1007%2FBF00218307

I think that AB-Pas is the best choice.You also can try the Best's carmine technique for neutral mucins or combination this carmine stain with AB.This combinated staining will demonstrate the neutral and acid mucins togather.

I agree with Sayed Mohsen. Moreover I think that you could use also High Iron Diamine tecnique (Spicer, 1965).
The high iron diamine method of Spicer is a useful technique for the detection of the highly acidic sulfomucins. This technique is selective for carbohydrates carrying a high negative charge density due to ionized sulfate groups. Hyaluronic acid and sialomucins are not demonstrated by this technique (Spicer 1965; Gad & Sylven 1969). When combined with the alcian blue protocol the high iron diamine technique facilitates the differentiation of sulfomucins from sialomucins in tissue sections.
When used together with the standard alcian blue procedure, sulfomucins and proteoglycans stain brown to black while sialomucins and hyaluronic acid stain blue.
Try Azocarmine Azan staining. this is a trichrome stain and it is very specific in that it stains mucous and mucus blue. Basal membranes also stain blue. Nuclear material stain red/purple red, muscle blue and red ( resulting in purple), chitin stains bright orange, the recipe is in Heidenheins text book or alternatively contact me and I will copy and send it to you

Toluidine blue 0,1% is a very fast and reliable method to stain mucous cells. There are a lot of variations of this method which particularly good and fast if applied to material embedded in historesins (glycol methacrylate). It will stain the mucus metachromatically in pink and the tissue will remain blue.

I've observed the acid mucopolysachharide from the intestine of scallop Pecten maximus by using Alcian Blue (Mowry,1956; Gave, 1968) and it's worked very well.
