I am going to do analysis of antioxidant activity of some medicinal plants, for that I should go through solvent extraction. I want to ask that should I use the same solvents for thin layer chromatography which are used for extraction of the plant material. Please help me out.
Definately you can use the same solvents for TLC. And it will be more easy if you can tell the solvents using and polarity of the product.

First of all, solvents are selected based on the constituents of the extracts which you will get after extraction. For knowing the chemical composition, you will have to perform phytochemical testing. Once you know the different chemical categories present in the extracts, you have to select the solvent systems for that particular chemical categories like alkaloids, flavonoids, phenolics etc. And you will have to do lots of hits and trials to get the best solvent system which gives you the best resolution and best separation. I think you need to have a thorough reading about the TLC and its principles, solvents and principles involved and detailed methodology. I am attaching some papers which will definitely clear all your doubts in this regard. I would also suggest you to read Wagner and Bladt's TLC atlas for all queries related to TLC. Of course.... you can use same solvent, as used in TLC, in your experiment.
@Gorentla sir and Prosenjit sir, I am using Acetone as polar solvent and Hexane or Ethyl acetate as non-polar solvent.
No problem, you can use any combination that should be convenient for you to extract or isolate the required product selectively. It is more familiar to the person handling the reaction and based on the nature of the product and impurities formed during the reaction.
You can use the same solvents in both your experiments and TLC.
Recommended mobile phases:
-DCM-MeOH (40:1, 30:1, 20:1, 10:1, 5:1)
-EtOAc-petroleum (3:1, 2:1, 1:1, 1:2, 1:3)
According to the retention factor of your samples, you will be able to decide which mobile phase suits you best.
