Which plant extract is better for antimicrobial sensitivity testing (disk diffusion assay)? centrifuged or non-centrifuged extract?
I think it depends on the active materials exist in either fractions against either of tested microorganisms, thus I suggest you primarily to try the both cases then you will see which one is better? Also in these cases, the solubility of the active materials is crucial factor, then the type of the solvent is a key factor for answering your question

Usually whole Crude extracts are used for the disc diffusion method due to myriad of chemical constituents present in the extract which yields higher efficacy. Incase of centrifugation process the solvents used to extract contains chemical compounds which are dissolved in it. So it depends on the experiment and chemical constituents you needed to test ?.

As per the Prof. Khaled Khleifat you have to try both the methods primarily to standardize the best one. I think both answers given above are best of knowledge.

In my opinion, the non ceterfuged is best .because in centrifugation may lose some materials.
In my opinion, non-centrifugal is best to retain the effect of the chemical effect of the plant extract in the sensitivity test.
