Personaly, I used methanol to extraxt phospholipids from lipid samples coming from meat. So, I suppose it will also be the best solvent for your method.

Chloroform:methanol (2:1 v/v) or toluene are perfect solvents for phospholipids, but you will have to test them in your particular application given that they evaporate at very different rates.

Typically phospholipids have some polarity, thus, I would use methanol. Neutral lipids are typically soluble in chloroform and glycolipids are soluble in acetone or a 9-1 mixture of acetone and MeOH.

We have used 'Hexipa' (hexane/2-propanol (3/2, by vol)) in our lab for years, no problem. All the above mentioned phospholipids plus a range of neutral phospholipids dissolves readily in Hexipa.

Methanol is best for phospholipids because of the polar charges that exist. Texan and chloroform are best for non polar neutral lipids.

Chloroform:methanol:water (5.5 : 5.5 : 5 v/v) can extract for most of cellular lipids (about 90% based on radioactive labeling data).  With 2% acetic acid in methanol is better for lipid extraction. For inositol phosphlipids, 1% HCl in methanol is better.

CERTAINLY, CHLOROFORM: methanol in 2:1 ratio

Uno roder to extract total lipids, I would agree with chloroform-MeOH (2:1 v/v).  This is the solvent to extract total lipids in the Folch and Dryer technique. However in order to separate lipids by silicic acid chromatography, technique, after total lipids are applied to the silicic acid columns, neutral lipids are washed off the column with chloroform. Phospholipids are then washed off the columns with MeOH. Finally, glycolipids are washed from the columns with either acetone or a 9:1 mixture of acetone & MeOH.

A combination of Chloroform and Methanol at the ratio of 2:1 would be better for extracting phospholipids (Folch method) .

Hexan is normally used for extracting NL. If you want to get pure PL, then first use hexan, that will de-oil your samples and will remove NL. I use ethanol to extract fish phospholipid from deoild sample. Hope, it will help. Cheers
