Hello,

I have a microscope slide with brain tissue on it. The tissue has been labeled with the fluorescent protein Dapi. In addition, the brain tissue was initially implanted with tumor cells transfected with mCherry fluorescent protein. The goal of this experiment is to see the cancer cells (mCherry) under fluorescence. So I want to scan the slide with an epifluorescent microscope. For this, I use one filter for Dapi and the other filter for mCherry is DsRed.

I want to know if the « Fluorescent mounting medium » provided by Dako company (distributed by Agilent) is a good choice for this experiment (https://www.agilent.com/cs/library/msds/SDS210_NAEnglish.pdf).

I have heard that the choice of the mounting medium can have a significant effect on fluorescence.

If the answer is no, what is the best choice to have optimal fluorescence for the DsRed filter and see mCherry fluorescent cells?

Thank you very much!

Laurence
