PRRSV infects macrophages and it found in pulmonary alveolar macrophages in lungs. And Macrophages presents in blood cells. The RNA isolated from white blood cells contains PRRSV RNA.
And Is PRRSV present in serum. If I isolate RNA from serum is their any possibilities of getting PRRSV RNA.
Now i have to start collection of samples from pigs. Please let me know which sample is good blood or serum.
Dear Phani , Good luck in your research and experiments as well , I suggest you to isolate the viral RNA from serum or tissue fluids (Lung) as recommended by OIE Manual (attached).
For collecting PRRSV , you do the isolation from serum since whole blood may interfere with your viral RNA isolation.another option is that isolate Peripheral blood nuclear cells by histopaq(sigma) from whole blood and isolate viral RNA. There are few kits are readily available for isolating total viral RNA from serum(150-200ul).

Whole blood also be included to extract RNA by trizol Method, in this method aquase phase containing clear and pure RNA, same time PCR inhibitory component from RBC present in the protein phase. So, in this way you can get the viral RNA from serum, WBC and RBC. Best of luck.
