Our work involves the mass extraction of protein from a fermentation broth and subsequent treatment of the proteins with acetone. We have not seen anything that will resuspend the proteins. This may be because the protein has been denatured and no longer has all the materials needed to go this route as these compounds remain in the solvent.
EMSA stands for Electrophoretic mobility shift assay. I am working on leaf proteins (nuclear) and studying DNA protein interaction. In many of the protocols, the precipitation is carried out by isopropanol and the pellet is being resuspended. I have modified the precipitation and involves washes with acetone. I need a suitable buffer to resuspend the pellet such that the native structure is retained. Thanks.
