Have you ever tested different polymerases for their suitability for multiplex PCR?
What's your opinion? Is it worth to buy those more expensive brands? Considering the budget of my project I have to find something that works well but is not too expensive.
expensive taq DNA polymerase are not always necessary sir, i have worked with 3-4 different types of Taq DNA polymerses on Y-STR markers, which gave the results with same intensities.
Taq polymerase makes a huge difference when multiplexing. I have tested at least 8 different Taqs in side by side experiments and two performed the best. I will add that Promega's taq was one of the two. I am doing large pool, medium, and small pool PCR experiments so the Taq must be pretty robust. Hope that this helps. I will add that the buffer system had to be tweaked a bit to get optimal results.
